Doody Review Services Reviewer: Gregory Reed, PhD (University of Kansas Medical Center) Description: This book pulls together relevant aspects of analytical chemistry, biochemistry and molecular biology, immunology, pharmacology and toxicology, and physiology as they apply to the study of drug disposition. Purpose: Effective planning, execution, and interpretation of studies in drug disposition requires an appropriate understanding of several separate areas of science. In the past this broad training was accomplished by following directions from senior investigators and by heeding their advice, while at the same time trying to recall principles taught didactically in very different contexts. This book aims to accelerate the training process by assembling the key theoretical and practical aspects of many areas and then stressing their application to the study of drug disposition. This is a highly worthy goal, and one that the book generally meets. Audience: The book is proposed as a text for senior undergraduate and graduate students, primarily those enrolled in pharmaceutical and chemical science programs. This audience could well be broadened to include post-docs and professionals in related fields, and even to experienced practitioners as a refresher course. Features: The first half of the book focuses on bioanalysis and on the use of the resultant data for pharmacokinetic and pharmacodynamic modeling. These chapters are quite straightforward in their ordering and their individual presentations. The subsequent sections discuss phase I and II enzymes and metabolism, in vitro approaches in drug metabolism, and the role of molecular biology in the field. These chapters, especially the metabolism and the molecular biology chapters, are somewhat problematic. The scope of the book requires that these be simplified and rather minimalist presentations of these areas, but this restriction sometimes lessens the clarity and, on a few occasions, the accuracy of what is presented. Assessment: The potential value of a single-source teaching and reference book for bioanalysis and drug metabolism, as opposed to having to mine relevant sections of numerous separate book, is great. This book is a worthy attempt. For a more detailed and broader treatment strictly of bioanalysis, Venn's Principles and Practice of Bioanalysis (Taylor & Francis, 2000) is preferred, but that book lacks the extensions to both the molecular side (enzymes, molecular biology) and to the whole organism side (pharmacokinetics, pharmacodynamics) that this book provides. |
