Nikon super resolution microscope research systems

In December 2009 Nikon Instruments announced the introduction of super resolution microscopes for N-SIM and N-STORM. Nikon's two new fluorescence microscopy technologies greatly enhance the spatial resolution of conventional optical microscopes, making it possible to view microstructures and nanostructures of living cells with molecular-scale resolution beyond the limits of optical microscopy.
N-STORM provides dramatically enhanced resolution that is more than 10 times greater than that of conventional optical microscopes, with "Stochastic Optical Reconstruction Microscopy" Storm not only increases the resolution in XY but also has the potential of increasing the Z resolution up to 50nm.
N-SIM is a new technology that uses stripe-pattern illumination and utilizes the “moiré effect” for creating super resolution images by image processing. The resulting optical resolution becomes twice as good as the resolution predicted using the N.A. of the objective lens.
In this presentation both techniques will be described and an overview of the Nikon N-SIM and N-STORM microscope systems will be given.

HIV and atherosclerosis: an unexpected connection

HIV disease is known to affect the immune system of infected subjects. These patients are also known to be at high risk for developing atherosclerosis but whether this risk is due to HIV infection or anti-HIV treatment remains controversial.
Our studies demonstrated a direct connection between HIV-1 replication and impairment of cholesterol metabolism. We found that HIV-1 protein Nef impairs activity of ABCA1, the main cellular cholesterol transporter, in HIV-infected human macrophages and affects reverse cholesterol transport in SIV-infected monkeys, and HIV-1 infection reduces levels of high density lipoprotein (HDL) in HIV-infected humanized mice.
We also demonstrate that pharmacological agents, such as agonists of the liver X receptor, that stimulate ABCA1 expression and reverse cholesterol transport exert potent anti-HIV activity and also reverse the HIV effect on HDL formation. Other lab projects include studies of a novel pro-inflammatory factor, extracellular cyclophilin, and genome-wide analysis of cellular factors involved in the pre-integration steps of HIV-1 replication.

Microtubule motors in vivo: moving on moving tracks

Cytoplasm organization and polarity of eukaryotic cells is largely defined by microtubule cytoskeleton and microtubule motors. Microtubule motors typically move cargo along microtubules in a bidirectional (saltatory) fashion. The goal of our work is to understand how multiple motors of opposite polarity transport cargo along microtubules, how they interact with each other, and how motors are regulated by associated proteins.
More recently, we discovered that a classical organelle motor, conventional kinesin, has a second important function, sliding of cytoplasmic microtubules against each other. We will discussed the mechanisms of microtubule sliding by kinesin and the role this sliding plays in generation of asymmetrical cell shape and formation of cellular processes.

Визуализация и анализ структуры активаторных хроматиновых блоков в индивидуальных хромосомных локусах

Активаторными хроматиновыми блоками называют комплексы регуляторных элементов, обеспечивающие активацию экспрессиии тех или иных генов или групп генов. Входящие в состав комплекса регуляторные элементы могут находиться на большом расстоянии друг от друга на молекуле ДНК. Соответственно, объединение этих элементов в общий активаторный комплекс возможно лишь при условии выпетливания разделяющих их сегментов хроматиновой фибриллы. Модель активаторных хроматиновых комплексов была предложена Де Лаатом и Гросвельдом на основании изучения пространственной организации локуса бета-глобиновых генов мыши с использованием техники фиксации конформации хромосомы (3С). К сожалению, техника фиксации конформации хромосомы имеет ряд существенных ограничений. Прежде всего, эта техника позволяет оценивать лишь относительные вероятности колокализаций различных пар геномных элементов. Колокализация трех и более геномных элементов в принципе не может быть продемонстрирована с помощью техники 3С. Далее, техника 3С не позволяет оценить долю клеток в популяции, в геноме которых один или оба исследуемых локуса имеют ту или иную пространственную конфигурацию. Причина этого заключается в том, что техника 3С вообще не детектирует линейную конфигурацию локуса.
В силу перечисленных ограничений метода 3С популярная модель активаторных хромтиновых комплексов до настоящего времени оставалась гипотетитческой. Совместно с лабораторией А.Б. Четверина (Институт белка РАН) мы разработали новый подход для анализа структуры активаторных хроматиновых комплексов, который получил сокращенное название INGRID (In Gel Replication of Interacting DNA). Принципиальным отличием данной процедуры является то, что анализируемые геномные элементы и комплексы таких элементов преобразуются в простые и смешанные молекулярные колонии, которые потом могут быть визуализированы. Число колоний различных типов может быть прямо подсчитано. Используя технику INGRID мы впервые продемонстрировали, что многокомпонентные активаторные комплексы существуют и число их значительно превосходит число двукомпонентных комплексов.

Морфогенез и его регуляция в культуре эпидермальных клеток человека

В последние несколько десятилетий получено множество новых данных относительно регенерации и морфогенеза у многоклеточных организмов. Одним из инструментов, позволивших сделать это, явилось развитие техники культивирования клеток и тканей. Эпидермис был одной из первых тканей, которые начали культивировать in vitro. Способность эпидермальных клеток к реконструкции многослойного пласта с последующим встраиванием в нормальные ткани после пересадки позволила успешно применять культивированные кератиноциты для закрытия ожоговых дефектов кожи и ран. Одновременно был открыт путь к изучению особенностей биологии эпидермальных клеток. В лаборатории давно ведутся исследования в этом направлении. В докладе речь пойдет о способности эпидермальных клеток к самоорганизации в ходе морфогенеза стратифицированного пласта, сохранении стволовых эпидермальных клеток в культуре, эпителио-мезенхимных взаимодействиях. Последние будут рассмотрены на примере взаимодействий клеток в волосяном фолликуле, биологии которого будет также посвящена некоторая часть доклада. Выяснилось, что волосяной фолликул содержит несколько популяций стволовых клеток, имеющих разное происхождение. Потенциал этих клеток кажется сегодня настолько широким, что кожу можно рассматривать в качестве весьма перспективного источника клеточного материала для регенеративной медицины. В ходе морфогенеза волосяного фолликула реализуется сложная цепь эпителио-мезенхимных взаимодействий. Волосяной фолликул – это уникальный орган млекопитающих, способный к многократной физиологической регенерации. Возможность воспроизведения в культуре начальных этапов морфогенеза волосяного фолликула дает возможность изучения его развития и патологий у человека.

Функциональный гетерохроматин и механизмы его сборки

Применение новых технологий в оценке транскриптома показало, что практически весь геном эукариот транскрибируется. Показана роль транскрипции и “коротких” РНК в формировании конститутивного центромерного гетерохроматина эукариот (дрожжи, растения). Будут рассмотрены также представления о роли транскрипции гетерохроматина в обеспечении стабильности генома и подавлении активности транспозонов. Указания на роль транскрипции сателлитных ДНК при формировании гетерохроматина у млекопитающих также будут представлены.

How to squeeze more out of your confocal microscope?

When talking about modern confocal systems the whole optical system from objective lens up to detector has to be considered. Focusing on the detailed specification of one of the components of the microscope setup cannot explain the overall specifications of the whole confocal system. Modern applications for confocal microscopes require fast or deep imaging which require a very efficient optical system. For combining fast and deep imaging the ultimate selection of specifications is needed.

Война и разоружение в мире вирусов и клеток

Вызываемые вирусами повреждения клетки (цитопатический эффект) обычно объясняют тем, что вирусы «приватизируют» клеточные ресурсы и используют их для собственной репродукции. Однако, накапливаются данные о том, что вирус-индуцируемая патология клетки может быть обусловлена, главным образом, противостоянием клеточных защитных и вирусных противо-защитных активностей, не имеющих прямого отношения к механизмам вирусного размножения. Важную роль в этом противостоянии играют механизмы альтруистического самоубийства клетки (апоптоз, некроз и, возможно, другие проявления программируемой гибели). Взаимное разоружение клетки и вируса может приводить к ослаблению клеточной патологии без существенного влияния на уровень репродукции вируса. Такое подавление может представлять собой новую стратегию борьбы с вирусными заболеваниями.

Morphogenesis & Antigenicity of HIV and Hepatitis Viruses

Since the last years, my team has concentrated on elucidating clue steps in viral assembly using various models including the hepatitis C virus (HCV), the hepatitis B virus (HBV) and, to a lesser extand, the human immunodeficiency virus (HIV). One long-term objective of our team is to take benefit of our expertise in viral morphogenesis to design vaccine strategies based on virus-like particles.
Before 2005, no effective cell-culture system for HCV was available, prompting us to develop an alternative model based on the production of HCV-like particles. Despite the development in 2005 of the first cell culture system supporting the complete HCV life cycle (model of the JFH-1 strain propagated in Huh7 cells), our model still remains unique to visualize the viral particle assembly by electron microscopy (EM). Thus, we showed that maturation of the HCV core protein by a cellular enzyme named signal peptide peptidase (SPP), which cleaves the core protein within its transmembrane domain, is a prerequisite for viral assembly. Cleavage of the core protein by SPP allows the assembly of the HCV-like particle and promotes trafficking of the core to the surface of lipid droplets. Many studies, conducted with the JFH1 strain, have further shown that core/lipid droplets association is essential for viral particles production. Using a 3D EM approach based on ultrathin serial sections, our model enabled to visualize this phenomenon. HCV-like particles were shown to assemble at the endoplasmic reticulum (ER) membranes distant but closely associated to the membrane monolayer surrounding the lipid droplets. Our work on HCV assembly focuses on the link between the virus and lipid droplets, which seems to be specifically supported by the HCV core protein. We set up a novel EM method for the morphometric analysis of the lipid droplets and used it to demonstrate that the production of the HCV core protein increased the number and size of lipid droplets. Moreover, a residue specific to genotype 3 viruses (the phenylalanine in position 164) amplified significantly this phenomenon. This observation allowed us to propose an in vitro model of the more severe liver steatosis observed in patients chronically infected by a genotype 3. More recently, we have analyzed the lipid droplets clustering induced by HCV, using different cell imaging approaches. We showed that this clustering resulted more likely from a neo-synthesis of lipid droplets in the ER membranes enriched in HCV core protein, rather than a simple redistribution of these organelles initially dispersed through the cell. Again, this original observation was important for understanding the mechanisms involved in virus-induced steatosis.
By adapting protocols developed for other RNA+ viruses, we have shown that host cell membranes harbouring HCV replication complexes contained also markers of autophagic vesicles. As in the case of the coronavirus involved in the SARS (Severe Acute Respiratory Syndrome), HCV appeared to induce autophagosome-like vesicles (also called DMVs for double-membrane vesicles), which could isolate the viral RNA from the cell machinery, thereby allowing the virus to escape from the innate cellular response.
The mechanisms of HBV subviral envelope particles morphogenesis remain unknown, but the S envelope protein was clearly identified as necessary and sufficient to induce budding (in vitro, the mass production of these S envelope particles is also the basis of the vaccine against hepatitis B for over 30 years). We recently showed by confocal and EM microscopy that the S protein self-assembles into filamentous particles in the ER. These filaments are packed in crystalline structures in the perinuclear area, and then transported to large cisternae related to the ERGIC to be converted into spherical particles. These particles are finally secreted outside the cell. Beyond the fundamental interest of these discoveries, our model easily allowed to identify by EM the capacity of assembly of S mutant proteins or S proteins fused to another protein. This prompted us to use this model to study the capacity of assembly of fusion proteins involving the HBV S protein and the HCV E1 and E2 envelope proteins. We showed that chimeric proteins in which the N-terminal transmembrane domain (TMD) was replaced by the TMD of E1 or E2 were competent for assembly, enabling the production of viral particles containing the complete HCV E1 and E2 envelope proteins. This strategy avoid the problem encountered in the production (secretion and purification) of HCV envelope proteins for a vaccine perspective. Interestingly, our vaccine candidate could be produced with the same industrial procedures used for the hepatitis B vaccine, and could potentially be a bivalent vaccine candidate against HCV and HBV.

Inhibitory signalling to the Arp2/3 complex steers cell migration

Cell migration requires the generation of branched actin networks that power the protrusion of the plasma membrane in lamellipodia. The Arp2/3 complex is the molecular machine that nucleates these branched actin networks. This machine is activated at the leading edge of migrating cells by the WAVE complex. The WAVE complex is itself directly activated by the small GTPase Rac, which induces lamellipodia. However, how cells regulate the directionality of migration is poorly understood. Here we identify a novel protein that inhibits the Arp2/3 complex in vitro, Arpin, and show that Rac signalling recruits and activates Arpin at the lamellipodial tip, like WAVE. Consistently, upon depletion of the inhibitory Arpin, lamellipodia protrude faster and cells migrate faster. The major role of this inhibitory circuit, however, is to control directional persistence of migration. Indeed, Arpin depletion in both mammalian cells and Dictyostelium discoideum amoeba resulted in straighter trajectories, whereas Arpin microinjection in fish keratocytes, one of the most persistent systems of cell migration, induced these cells to turn. The existence of a Rac-Arpin-Arp2/3 inhibitory circuit embedded within the positive feedback loop that maintains directional migration can account for this conserved role of Arpin in steering cell migration. Conversely, loss of this inhibitory circuit renders the positive feedback loop unrestrained and might commit carcinoma cells to the invasive state.

Dynamics of Polycomb chromatin domains under conditions of increased molecular crowding

A Polycomb (PcG) body is an orphan nuclear subcompartment characterized by accumulations of Polycomb repressive complex 1 (PRC1) proteins. However, seemingly contradictory reports have appeared that describe the PcG bodies either as protein-based bodies in the interchromatin compartment or chromatin domains. In this respect, molecular crowding is an important factor for the assembly and stability of nuclear subcompartments. In order to settle this contradiction, crowding experiments, that represent a convenient model distinguishing between interchromatin and chromatin compartments, were carried out.
In sucrose-hypertonically induced crowding, we observed in U-2 OS cells that PcG bodies disappeared, but persisted as nuclear domains characterized by accumulations of DNA. This phenomenon was also observed in cells hypertonically treated with sorbitol and NaCl. Importantly, the observed changes were quickly reversible after re-incubation of cells in normal medium. We found that the PcG foci disappearance and the dissociation of PRC1 proteins (BMI1 and RING1a proteins) from chromatin was associated with their hyper-phosphorylation. In addition, under hyper- and hypotonic conditions, the behaviour of the PcG bodies differed from that of typical nucleoplasmic bodies. We conclude that PRC1 proteins accumulations do not represent a genuine nuclear subcompartment. The PcG body is a chromatin domain, rather than a nucleoplasmic body.

Цитоскелет как система путей внутриклеточного транспорта в клетках животных


От внутриклеточного транспорта зависят многие процессы, жизненно важные как для отдельной клетки, так и для организма в целом. Так, например, эндо- и экзоцитоз, существование которых невозможно без такого транспорта, обеспечивают на уровне организма работу желёз, макрофагов и нейронных сетей. Отвечая за процессы клеточной поляризации и локомоции, внутриклеточный транспорт обеспечивает таким образом заживление ран, развитие воспалительных процессов, рост сосудов и нормальное протекание эмбриогенеза.
Внутриклеточный транспорт осуществляется моторными белками, которые перемещают органеллы вдоль элементов цитоскелета – микротрубочек и актиновых филаментов. Таким образом, в клетках существуют две транспортные сети, актиновая и тубулиновая. В результате многочисленных исследований в данной области были расшифрованы структура моторных белков и белков цитоскелета, проведена 3D-реконструкция молекул кинезинов, динеинов и миозинов и описаны механизмы перемещения моторов по соответствующим филаментам. Однако ряд фундаментальных вопросов об организации транспорта оставался без ответа, не давая возможности объединить все известные факты в одну непротиворечивую схему.
Полученные нами данные позволили существенно углубить имевшиеся представления об архитектуре тубулиновой транспортной системы. В частности, был открыт и детально охарактеризован саморегулирующийся механизм позиционирования центросомы в интерфазной клетке; впервые изучена роль двух новых белков центросомы в процессе удержания на ней микротрубочек и в обеспечении радиальности их расположения в цитоплазме. Кроме того, были внесены серьёзные изменения в представления о том, какие параметры транспорта обусловлены свойствами самой транспортной сети и составляющих её элементов цитоскелета. В частности, был изучен вопрос о механизмах миозин-зависимого транспорта по актину и о влиянии динамики микрофиламентов на направленность этого транспорта.
Понимание принципов организации транспортных систем клетки позволяет в деталях исследовать многие процессы, интересные как с точки зрения фундаментальной цитологии, так и прикладных биомедицинских исследований. Это позволяет говорить о том, что полученные данные имеют не только теоретическое, но и практическое значение.

Анализ роли канонического сигнального каскада NFkappaB в эмбриональном развитии


Канонический сигнальный каскад NFkappaB регулирует иммунитет, воспаление, организацию цитоскелета, клеточную адгезию и ингибирует апоптоз связанный с развитием раковых опухолей. Его активация в основном зависит от протеинкиназы Ikk2 (Ikk-beta), которая фосфорилируя негативный регулятор NFkappaB - IkappaB запускает множество процессов и в том числе транскрипцию генов регулирующих развитие различных клеток и органов. Исследование роли Ikk2 в развитии осложнено тем, что панэмриональный нокаут Ikk2 у мышей прерывает эмбриогенез вследствие апоптоза в печени. Следовательно роль этого гена в развитии млекопитающих иучали в основном in vitro или в ткане-специфичных мутантах. Клеточные события регулируемые Ikk2-зависимым NFkappaB зависят от микротрубочек. Дефицит Ikk2 снижает экспрессию VEGF и увеличивает проницаемость капилляров. Для того чтобы проанализировать роль Ikk2 в развитии данио (Danio rerio B.) мы создали две различных мутации Ikk2. Их анализ и будет представлен на этом семинаре.

Functional organization of the genome and its epigenetic
state studied in engineered mammalian cell systems


Chromatin-mediated epigenetic gene regulation is crucial for cellular identity in higher eukaryotes. The epigenetic state plays an important role in directing gene expression variability, achieving long-lasting, unperturbed expression states. Epigenetic regulation can be dynamic, occurring as (transient, stochastic) oscillations in different cells in a clonal population (due to inherent ‘intrinsic’ noise). This dynamic nature explains the lack of conclusive understanding of epigenetic regulation since most observations assume cells are in phase and are based on cell-population averages. We use engineered mammalian cell systems allowing to modulate the (epi)genetic state at a single gene locus at predefined regions of the genome and measure input-output relationships of (epi)genetic regulation in multiple cells in a population. We developed techniques allowing systematic, quantitative measurements of changes in the transcription rate as function of the epigenetic chromatin state in single living mammalian cells with high precision and real-time and high-resolution microscopy combined with stochastic modeling-technology adding variable levels of epigenetic regulatory complexity. Our approach of modeling and quantitative sampling in designed mammalian cell systems is proof-of-principle to understand complex epigenetic regulatory behavior and opens an unexplored field of research with great potential for medicine.

Сравнительный анализ и перспективы современной микроскопии


Самое досадное в световой микроскопии – это ограничение разрешения на самом интересном месте, поскольку длина волны видимого света сопоставима с размерами клеточных органелл. Практические границы латерального (XY) разрешения составляют около 200 нм (5 линий на микрон) для видимого света и идеальной оптики (Аббе). Это серьезное ограничение для биологии клетки, размер которой составляет около 10-20 мкм, то есть всего 50-100 линий разрешения. Таким образом, мы можем различить около 2,000-8,000 деталей в клетке среднего размера и всего около 20 - в бактерии. Однако, мы обычно мы не достигаем даже этих скромных цифр, особенно если критерий Никвиста для цифрового детектора не был соблюден. Несколько современных подходов позволяют обойти предел Аббе, но не бесплатно. Микроскопия в просвечивающем мягком рентгеновском диапазоне 2-4 нм позволяет изучать монослой клеток in vitro без разрезания и высушивания. Контраст возникает за счет поглощения атомами углерода. Улучшение разрешение с 200 до 20 нм позволяет различать практически все клеточные органеллы, включая рибосомы и микротрубочки. В настоящее время самым надежным источником мягкого рентгена является синхротрон. С другой стороны, рентгеновская оптика дорога и малоэффективна. С третьей, поле зрения составляет 20-30 микрон. Нам удалось найти принципиальное решение всех трех проблем

Структурно-функциональные различия цитоплазматических изоформ актина в немышечных клетках

Актины, особенно цитоплазматические или немышечные изоформы, играют ведущую роль в ключевых клеточных процессах, таких как адгезия, миграция, поляризация и деление клеток. Используя специфические моноклональные антитела мы исследовали распределение бета- и гамма- цитоплазматических актинов в фибробластах и эпителиальных клетках. С помощью лазерной конфокальной микроскопии мы показали, что бета-актин преимущественно локализован в филоподиях, стресс-фибриллах, кольцевых пучках и адгезионных межклеточных контактах, что предполагает роль этой изоформы в клеточной адгезии и сокращении. Напротив, гамма-актин организован по-разному в зависимости от клеточной активности: в виде кортикальных и ламеллиподиальных сетей в движущихся клетках, что предполагает его важную роль в клеточной подвижности; в стационарных клетках гамма-актин также может быть локализован в дорзальных стресс-фибриллах. Исследование функций изоформ было проведено с помощью специфического уменьшения экспрессии бета и гамма-актинов РНК-интерференционным методом. Клетки, трансфицированные малыми интерферирующими РНК, избирательно подавляющими экспрессию бета или гамма-актина, имели выраженные морфологические различия: при уменьшении экспрессии бета-актина клетки хорошо распластывались и теряли бета-актиновые пучки; напротив, при уменьшении экспрессии гамма-актина клетки приобретали сократимый фенотип с толстыми пучками актина без ламеллиподиальных структур. Более того, уменьшение экспрессии каждой из изоформ актина по-разному влияло на движение, подтверждая их специфические роли в клеточной подвижности. Дополнительные данные о различной роли изоформ актина в миграции клеток были получены при сравнительномо изучении нормальных и неопластически трансформированных и опухолевых клеток. Также получены данные о различной роли этих изоформ в клеточном делении. Наши результаты выявили принципиально новые аспекты организации изоформ актина, предполагающие их функциональные различия.

The actomyosin cytoskeleton: a master-regulator of intercellular junctions and epithelial differentiation


Adherens junctions (AJ) and tight junctions (TJ) are critical regulators of epithelial cell differentiation, establishment of apico-basal cell polarity and integrity of epithelial barriers. AJ/TJ disassembly mediates disruption of epithelial barriers during mucosal inflammation and drives epithelial to mesenchymal transition (EMT) and tumor metastasis. AJ and TJ are associated with the actomyosin cytoskeleton, which is essential for junctional integrity and dynamics. This talk will focus on how actin filaments and non-muscle myosin II motor control epithelial cell-cell adhesions and regulates barrier properties of epithelial monolayers. It will describe unique connections between cytoplasmic actin isoforms and specific epithelial junctional complexes, and will reveal inductions of the EMT-like cell phenotype after expressional down-regulation of cytoplasmic actins. Furthermore, this talk will discuss the roles of non-muscle myosin II motor in the remodeling of epithelial junctions in vitro and disruption of the intestinal barrier during mucosal inflammation in vivo.

Tropomyosin: The path from cell architecture and signalling to cancer therapy

All organisms from bacteria to plants and animals have evolved the capacity to generate multiple ‘actin’ filaments which differ in their composition and perform functionally distinct roles within the cell. All animals sort both their actin and tropomyosin isoforms to different locations within a single cell. This sorting often correlates with known specialised actin filament functions within the cell. Sorting is often more easily detected within differentiated cells. Sorting appears to be the result of local assembly and is maintained by the integrity of local filaments and higher order structures. Most recently we have identified a second tropomyosin associated with the Golgi and two tropomyosins almost exclusively located in the lamaellipodium. We have also found that 70-90% of phalloidin-positive pixels in stained cells are also positive for tropomyosin indicating that the majority of actin filaments in the cytoskeleton are likely to contain tropomyosin. Tropomyosin isoforms are engaged in a broad range of isoform-specific functions. Recent work has focussed on the cancerassociated tropomyosin Tm5NM1. All known tumour cells retain Tm5NM1 and cancer progression is usually associated with reductions in the levels of most Tm’s except Tm5NM1. Atomic Force Microscopy has been used to show that Tm5NM1 can increase cell stiffness to a level substantially greater than other isoforms. Mice which over-express Tm5NM1 display increases in the sizes of specific tissues and organs whereas the gene knockout has the converse result. The levels of Tm5NM1 regulate glucose uptake into tissues, possibly via the role of Tm5NM1-containing actin filaments in the exocyst complex. Previous studies have shown that tropomyosin isoforms can regulate the interaction of actin filaments with actin binding proteins and motors in an isoform specific manner and that tropomyosin is limiting for actin filament accumulation in cell culture. More recent data has shown in mice that tropomyosin supply is limiting for actin filament levels in vivo. In addition, the levels of Tm5NM1 can regulate the levels of exocyst components Sec8 and Myo1c in vivo. This correlates with previous work indicating that the steady state levels of tropomyosin isoforms can determine the steady state levels and activity of actin binding proteins and motors in cell culture. The cancer-associated Tm5NM1 is the only tropomyosin which promotes cell proliferation and this activity may relate to its impact on tissue and organ size. Mouse embryo fibroblasts which over-express or are knocked out for Tm5NM1 display reciprocal effects on cell proliferation. Knockout cells show reduced proliferation as a result of a defect in the canonical MAPK pathway. This is due to complete shutdown of ERKII regulation of cell proliferation. Recent data indicates that Tm5NM1 actin filaments are absolutely required for nuclearsignal-dependent nuclear translocation of ERKII. In addition, further data indicates that other products of the TPM3 gene are required for embryonic stem cell differentiation and the TPM4 gene is required for normal senescence. This suggests that tropomyosins may be able to regulate different signalling pathways in an isoform specific manner. We have recently published the development of anti-Tm5NM1 drugs for the treatment of cancer. Drug treatment leads to the disassembly of the tumour cell actin cytoskeleton but does not impact striated muscle or differentiated cells in culture.

"The role of nuclear organization in regulation of transcription and the occurence of chromosome translocations in human lymphomas"

Chromosomes are confined to chromosomal territories in the nuclear space. Translocations that lead to exchanges of chromosome arms may disrupt this cellular order and lead to relocalization of genes within the nucleus.
We have studied the localization of cyclin D1 (CCND1) and c-myc gene regions within the nuclei in normal human lymphocytes and the mantle cell lymphoma and Burkitt lymphoma cells and cell lines. We have shown that both CCND1 and c-myc genes were relocalized form the chromosomal periphery into the central perinucleolar region following translocation. We have also shown the presence of nucleolin-binding sites in the vicinity of the CCND1 gene and show that they upregulate transcription from the CCND1 promoter. We also show intensive binding of nucleolin to the CCND1 gene promoter after the translocation and also show that the chromatin organization of the translocated regions changes after the translocation and that these changes span over very large regions. The same pattern is observed in the Burkitt lymphoma, where c-myc is upregulated by nucleolin.
We propose a novel mechanism of carcinogenesis brought about by chromosomal translocations where the relocalization of chromosomal territories within the nuclear space leads to misregulation of translocated genes. Similar mechanisms may be involved in the genesis of translocations in human B-lymphocytes.

Оптимизация фибринолитической терапии плеврального фиброза – от изучения механизмов in vitro и ex vivo и in vivo моделей на животных, к лечению эмпиемы.

Фиброз плевральной полости является одним из осложнени пневмонии, операций на сердце и онкологических заболеваний. Дисрегуляция фибриногенеза и подавление эндогенной фибринолитической активности приводит к аккумуляции фибрина и накоплению плевральной жидкости (гноя), которые механически воздействуют на легкое, нарушая процесс дыхания. Плевральный фиброз сопровождается болевым синдромом и часто приводит к сепсису. Несмотря на успехи современной медицины, частота локулированных плеврального фиброза не падает, а их летальность достигает 20%. Фибринолитическая терапия (ФТ) активирует эндогенный фибринолиз и позволяет минимизировать или избежать хирургического вмешательства.
Для выяснения молекулярных механизмов, контролирующих развитие фиброза и влияющих на клинический результат ФТ с использованием проурокиназы и тканевого активатора плазминогена мы использовали in vitro, ex vivo и in vivo (модели плеврального фиброза у кроликов) подходы, а также современные методы диагностики (ультразвуковая сонография (УЗИ), компьютерная (КТ) и магнитно-резонансная (МРТ) томография). Изучение механизма процессинга проурокиназы in vivo позволил определить время фибринолиза в плевральной полости (6-8ч), что было подтверждено результатами УЗИ и КТ. Анализ плевральной жидкости животных и человека показал, что активная форма ингибитора активаторов плазминогена (ПАИ-1) является биомаркером и молекулярной мишенью при ФТ. 1000 кратное уменьшение скорости реакции между протеиназой и ПАИ-1 оказолось неэффективной in vivo. Использование антител, «превращающих» ПАИ-1 из «суицидного» ингибитора в субстрат для протеиназ и увеличивающих «стехиометрию ингибирования», позволило уменьшить эффективную дозу проурокиназы в 8 раз. Изучение инфекционной модели плеврального фиброза у кроликов и анализ образцов плевральной жидкости больных выявило общие параметры: увеличение активного ПАИ-1, внеклеточной ДНК, и наличие «фибринолитического потенциала», которые могут быть использованы для уточнения диагноза, персонализации дозировки и прогноза фибринолитической терапии. Нами был предложен молекулярный механизм внутриплеврального фибринолиза при ФТ. Высокие уровни активного ПАИ-1 и внеклеточной ДНК, относительно низкие скорость внутриплеврального фибринолиза и индивидуальные различия в эдогенной фибринолитической системе рассматриваются как основные факторы, влияющие на клинический исход фибринолитической терапии плеврального фибрроза. Механизм положен в основу оптимизации фибринолитической терапии в моделях инфекционной эмпиемы у кроликов и анализа человеческих образцов, полученных в ходе клинических испытаний.

Correlative Microscopy: to discover life-changing answers faster

Biological systems are vastly complex, but the ability to see detailed relationships between structure and function at various levels of resolution and within a functional correlated context brings greater understanding. Correlative light and electron microscopy (CLEM) provides labeling for fluorescence microscopy, allowing the study of dynamic events in living cells. It also gives access to high resolution data collected from electron microscopy. This approach has been used in an increasing number of scientific publications and has proved to be a valuable tool for researchers in different fields of life science. I will tell about latest Correlative Microscopy solutions. You’ll see examples of CLEM workflow that fulfill the varying needs of cell biologists, from imaging of living cultured cells to 3D analysis of cellular ultrastructure.
