[править] Clean the Enricher

IMPORTANT! Clean the Enricher after a run. Cleaning the Enricher after every run removes residual beads from the instrument. If a run was performed overnight or over the weekend, clean the Enricher as soon as possible within a 3-day period.

  1. Disconnect the tubing from the listed components below, then reconnect the tubing to the Cleaning Manifold (see Table). Make sure that tubing connections are tightly secured to prevent leaks.

    •SOLiD™ EZ Bead™ Emulsifier tubing, top

    IMPORTANT! Do not disconnect the tubing from: Butanol bottle, Reagent racks, Waste bottle

  2. Remove and dispose all disconnected tubes, columns, and the Enricher Filter from the Enricher.
  3. Empty the waste bottle, then return the bottle to the Enricher
  4. Start the cleaning after a run:
    a. Ensure that the green «POWER» on the front panel is lit. If the green «POWER» status is not lit, toggle the power switch to the on position on the back instrument panel, wait until the current run is complete.
    b. Open the Enricher software by double-clicking the icon. The Enricher control window displays.
    c. Click the scale-selection button to select Clean in the control window.
    d. Click the start-run button. A checklist displays.
    e. Complete and check off each item in the checklist. You cannot proceed with cleaning until all of the steps are checked off. You can exit the checklist by clicking the stop button. After you complete the checklist, ensure that the Enricher doors are closed. Click the check button at the bottom of the checklist to begin cleaning. The yellow «IN USE» on the front status panel is lit.
    Cleaning the instrument takes ~1.5hours. After cleaning is complete, the Emulsifier stops automatically. The green «POWER» status on the front panel is lit.
    f. Close the software.
  5. Maintain the Enricher according to these steps:
    If you will be using the Enricher… Do the following…
    <3 days Leave the reagent racks on the instrument, and close the door to the instrument. Keep the tubing connected to the Cleaning Manifolds until the next sample run

[править] Prime 1: Prepare the Enricher before long-term storage

IMPORTANT! You must perform the cleaning protocol before priming the Enricher for long-term storage (more than three days).
  1. Ensure that all tubes, columns, and the Enricher Filter are removed from the Enricher.
  2. Fill the bottles on the cleaning racks with Nuclease-free Water (see Table 3 )

  3. Empty then reinstall the waste bottle.
  4. Install cleaning racks A and B with the bottles filled with water:
    a. Install each rack on the railing.
    b. Slide each of the two racks into the appropriate slot until each rack is slid fully into position, when the rack stops.
  5. Start the priming:
    a. Ensure that the green “POWER” on the front panel is lit. If the green “POWER” status is not lit, toggle the power switch to the on position
    on the back instrument panel, wait until the current run is complete.
    b. Open the Enricher software by double-clicking the icon. The Enricher control window displays.
    c. Click the scale-selection button to select Prime in the control window.
    d. Click the start-run button. A checklist displays.
    e. Complete and check off each item in the checklist. You cannot proceed with priming for long-term storage until all of the steps are checked off. You can exit the checklist by clicking the stop button. After you complete the checklist, ensure that the Enricher doors are closed. Click the check button at the bottom of the checklist to begin priming for long-term storage. The yellow “IN USE” on the front status panel is lit.
    Priming the instrument for long-term storage takes ~30minutes. After priming for long-term storage is complete, the Enricher stops automatically. The green “POWER” status on the front panel is lit.
  6. Close the software.
  7. Empty the waste bottle, then return the bottle to the Enricher.
  8. Leave the cleaning racks A and B on the instrument, and close the door to the instrument. Keep the tubing connected to the cleaning manifolds until the next sample run.
    IMPORTANT! You need to keep the cleaning racks and bottles for repeated use.
  9. Close the instrument doors.

[править] Prime 2: Prepare the Enricher for first-time use or after long-term storage

IMPORTANT! Do not disconnect the tubing from:Butanol bottle, Waste bottle
  1. Ensure that the tubing is connected to the cleaning manifold.
  2. Fill the bottles on the cleaning racks with Nuclease-free Water (see Table 4 )

  6. Empty the waste container, then reinstall it on the Enricher.
  7. To start a run on the Enricher, prepare and install reagents on the instrument(SOLiD™ EZ Bead™ Enricher).
